Freshwater microcrustacean Daphnia magna Straus as an early screen model to compare toxicity of acetylcholinesterase inhibitors

چکیده
Daphnia magna is a freshwater microcrustacean which is often used for acute and chronic toxicity testing in aquatic ecotoxicology. Recently, tests with daphnids have been used to prescreen the toxicity of newly synthesized acetylcholinesterase reactivators (oximes), which appear as weak inhibitors of acetylcholinesterase (AChE). In our study we investigated and compared the toxicity of five reversible acetylcholinesterase inhibitors, two of them non ionic (tacrine and 7-MEOTA) and three ionic (berberine chloride, 1-methyltacridium iodide and ethidium bromide). Tests were carried out according to the methodology described in the standard EN ISO 6341. EC50 values as well as dose-response curves were calculated for all tested inhibitors. In general, non ionic substances were found to be less toxic than ionic substances. Estimated EC50 values in tests taking respectively 24 h and 48 h were as follows: tacrine 3000 resp. 1515 μg.l, 7-MEOTA 3606 resp. 707.2 μg.l, berberine chloride 903.6 resp. 822.4 μg.l, 1methylacridinium iodide 714.3 resp. 233.5 μg.l and ethidium bromide 644.3 resp. 291.1 μg.l. The higher toxicity of 7-MEOTA in time compared to tacrine and the similar toxicity of berberine chloride in both tests are explained as a consequence of their metabolism in the daphnid’s body.

As a result of a widespread application in numerous industrial processes, chromium is a contaminant of many environmental systems. Chromium and their compounds are toxic to both invertebrates and vertebrates and, for this reason, there has been a search for suitable and less toxic alternatives. Molybdenum compounds have been studied as alternative to chromium compounds for some industrial appli...

Titanium dioxide nanoparticles (nTiO2) are the world's second most widely consumed nanomaterial and large quantities of this material enters the aquatic ecosystem annually. Therefore, understanding the effects of nTiO2 on aquatic organisms is very important. The present study used Daphnia magna as a model freshwater organism and Pontogammarus maeoticus as a brackish water organism to evaluate s...

While the commercialization of single-walled carbon nanotubes (SWCNTs) is rapidly expanding, the environmental impact of this nanomaterial is not well understood. Therefore, the present study evaluates the acute aquatic toxicity of SWCNTs towards two freshwater microalgae (Raphidocelis subcapitata and Chlorella vulgaris), a microcrustacean (Daphnia magna), and a fish (Oryzias latipes) based on ...

The Avoidance Responses of Daphnia magna to the Exposure of Organophosphorus Pesticides in an On-Line Biomonitoring System
In this study, avoidance behavior of the freshwater cladoceran Daphnia magna Straus was used as indicator to assess the early stress of accidental organophosphorus pesticide (OP) contamination. The movement behavior was detected by a multi-species biomonitoring system. There was obvious concentration–response relationship between the OP stress and the behavioral response even at sublethal expos...
